1)面筋蛋白是小麦的贮藏蛋白,不溶于水。它主要由麦胶蛋白(一种醇溶蛋白)和卖谷蛋白组成。

2)这两种蛋白分离:在稀酸中溶解面筋,添加乙醇配成70%的乙醇溶液,然后添加足够的碱以中和酸,在4℃下放置一夜,麦谷蛋白沉淀,溶液中剩下麦胶蛋白。麦胶蛋白(是一大类有类似特性的蛋白)水合时胶黏性极大,这类蛋白质抗延伸性小或无,其造成面团黏合性的原因。麦谷蛋白是一类不同组分的蛋白,多链,有弹性但无黏性,使面团具有延伸性。之所以面筋蛋白能彼此相互作用形成具有粘弹性的面团,是因为:

①面筋蛋白中Glu含量非常高,在蛋白以Gln的形式存在,且面筋蛋白中碱性aa含量少,Lys少,面筋蛋白基本没有潜在负电荷,仅有少量潜在正电荷,电荷密度小,所以蛋白中相互排斥里弱。

②面筋蛋白中蛋白水平高,其肽键不易弯曲,在蛋白链中,凡是在有蛋白存在的地方,都出现一个硬节,使蛋白不易形成a-螺旋。
